De plus, il cible les radicaux carboxyles alcianophiles (glycosyl\u00e9s) agissant comme glycoprot\u00e9ines et glycolipides transmembranaires. Le <\/span><\/span><\/span><\/span><br \/>\n<span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\">bleu alcian permet de visualiser les prot\u00e9oglycanes dans les tissus, notamment dans le cartilage. Il permet de mettre en \u00e9vidence la pr\u00e9sence de glycosaminoglycanes (GAG), composants essentiels de la matrice extracellulaire.<\/span><\/span><\/span><\/span><\/p>\n<h4><strong><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\">Application :<\/span><\/span><\/span><\/span><br \/>\n<\/strong><\/h4>\n<p><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\">L&#8217;\u00e9tude en microscopie \u00e9lectronique b\u00e9n\u00e9ficie consid\u00e9rablement de l&#8217;application du bleu alcian. Ce colorant est sp\u00e9cifiquement utilis\u00e9 pour visualiser les prot\u00e9oglycanes dans les tissus, en particulier le cartilage. Cette technique de coloration permet de mettre en \u00e9vidence la pr\u00e9sence de glycosaminoglycanes (GAG), composants essentiels de la matrice extracellulaire.<\/span><\/span><\/span><\/span><\/p>\n<ul>\n<li><strong><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\">M\u00e9thodologie :<\/span><\/span><\/span><\/span><br \/>\n<\/strong><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"> Le principe de concentration critique en \u00e9lectrolytes est appliqu\u00e9 lors de la coloration au bleu alcian. Le cartilage \u00e9piphysaire de rat est fix\u00e9 au glutarald\u00e9hyde. La coloration est r\u00e9alis\u00e9e \u00e0 pH 5,8 sans MgCl\u2082 et en pr\u00e9sence de concentrations variables de MgCl\u2082 (0,05, 0,4, 0,5, 0,9 et 1,0 molaire). La comparaison est effectu\u00e9e avec la coloration et la fixation simultan\u00e9es \u00e0 pH neutre. Les deux m\u00e9thodes permettent de colorer le mat\u00e9riel intracellulaire dans les vacuoles et le mat\u00e9riel matriciel non collag\u00e9nique.<\/span><\/span><\/span><\/span><\/li>\n<li><strong><strong><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\">Observations :<\/span><\/span><\/span><\/span><br \/>\n<\/strong><\/strong><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"><span style=\"vertical-align: inherit;\"> Le mat\u00e9riel color\u00e9 est constitu\u00e9 de granules arrondis ou polygonaux. Ces granules s&#8217;accumulent autour des cellules dans les zones prolif\u00e9ratives et hypertrophi\u00e9es du cartilage. La distribution et la structure du mat\u00e9riel matriciel positif au bleu alcian restent coh\u00e9rentes dans diff\u00e9rentes conditions de coloration. Une concentration accrue en MgCl\u2082 entra\u00eene une r\u00e9duction de la coloration au bleu alcian, sugg\u00e9rant que le sulfate de chondro\u00eftine est un composant majeur des granules matriciels.
