Recipe / Preparation : PBS (Phosphate Buffer Saline) 1X, pH 7.4.

To prepare  500 ml of PBS  (1X, pH 7.4)

Component Amount Concentration
Sodium chloride (mw: 58.44 g/mol) 4 g 0.137 M
Potassium Chloride (mw: 74.55 g/mol) 0.1 g    0.0027 M
Sodium Phosphate Dibasic (mw: 141.96 g/mol) 0.72 g        0.01 M
Potassium Phosphate Monobasic (mw: 136.09 g/mol)     0.1225 g 0.0018 M

 


  1.  Prepare 450 mL of distilled water in a suitable container.
  2. Add 4 g of Sodium chloride to the solution.
  3. Add 0.1 g of Potassium Chloride to the solution.
  4. Add 0.72 g of Sodium Phosphate Dibasic to the solution.
  5. Add 0.1225 g of Potassium Phosphate Monobasic to the solution.
  6. Adjust solution to desired pH ≈ 7.4 or other pH.
  7. Add distilled water until the volume is 500 ml
