Sorensen phosphate buffer, also known as mixed sodium phosphate salts, is widely used in biology. The osmolarity of a 0.1 M solution at pH 7.2 is 226 milliosmoles; with the addition of 0.18 M sucrose to thesolution, the osmolarity rises to 425 milliosmoles. pH can be specified when ordering. The maximum shelf life after receipt is 3 months. All products used are chosen for their purity. Storage: 4-8°C. Sorensen phosphate buffer is available in different pH ranges: 5.8 – 8.

Application :

Sorensen phosphate buffer serves as a physiologically relevant buffer widely utilized in various experiments. It closely mimics certain components found in extracellular fluids, rendering it non-toxic to cells. The buffer demonstrates minimal pH fluctuations with temperature shifts and remains stable for several weeks when stored at 4°C. However, it is prone to precipitation during fixation and tends to form precipitates with uranium acetate and react with lead salts. Additionally, it slowly contaminates with microorganisms.

Tip :

For optimal preparation, phosphate buffer solutions should be stored at 4-8°C, and the shelf life after opening is 3 months.

Mother Solutions:

  • Sodium dibasic phosphate 0.2 M 1 liter
  • Sodium monobasic phosphate 0.2 M 1 liter

Working Buffer: Prepare a 0.1 M working buffer using 100 ml of Sorensen phosphate buffer. Mix X ml of sodium dibasic phosphate 0.2 M with Y ml of sodium monobasic phosphate. Dilute to 100 ml with ddH20 or dilute 1:1 with fixative.

Osmolarity varies by adjusting the phosphate molarity or by adding sucrose, glucose, or sodium chloride. At pH 7.2:

  • 0.10 M = 226 mOs (milliosmoles)
  • 0.05 M = 118 mOs
  • 0.075 = 180 mOs
  • 0.15 M = 350 mOs
