Product description :

The PHEM buffer is primarily used for tissues and cell cultures undergoing processing for immunocytochemical immunolabeling studies. Most antigens, especially intracellular antigens, stain more effectively with PHEM buffer than with PBS buffer. It also has a more limited effect on biochemical reactions and enzymes. A significant advantage of using PHEM is its excellent preservation of cell ultrastructure, particularly of microtubules.

Antibodies are optimized by the immune system to function in the sodium-rich environment of the blood. However, the intracellular milieu is dominated by potassium, so efforts should be made to recreate these conditions. Buffers such as PHEM or CSK have been developed to be used in permeabilized and unfixed cells in function-based assays, and they can be used on fixed and permeabilized cells instead of PBS, particularly useful with cytoskeletal or cytoplasmic antigens.

In terms of signal intensity, approximately 10% of cellular antigens stain significantly better in PHEM buffer than in PBS, 50% stain moderately better in PHEM, and 30% show no difference. Therefore, trying both PHEM and PBS when using an antibody for the first time is advisable. There are also other buffers available for experimentation.
