Alcian Blue is used to visualize proteoglycans within tissues, particularly in cartilage. It helps highlight the presence of glycosaminoglycans (GAGs), which are essential components of the extracellular matrix.

  • Methodology :
    The critical electrolyte concentration principle is applied during Alcian Blue staining. Rat epiphyseal cartilage is fixed with glutaraldehyde. Staining is performed at pH 5.8 without MgCl₂ and in the presence of varying concentrations of MgCl₂ (0.05, 0.4, 0.5, 0.9, and 1.0 molar). Comparison is made with simultaneous staining and fixation at neutral pH. Both methods result in staining of intracellular material within vacuoles and non-collagenous matrix material.
  • Observations :
    Stained material consists of rounded or polygonal granules. These granules accumulate around cells in the proliferative and hypertrophied zones of cartilage. The distribution and structure of Alcian Blue-positive matrix material remain consistent across different staining conditions.
